Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.245C>A (p.Ser82Tyr), citing Ambry Variant Classification Scheme 2023: The p.S82Y variant (also known as c.245C>A), located in coding exon 2 of the DICER1 gene, results from a C to A substitution at nucleotide position 245. The serine at codon 82 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
