NM_004655.4(AXIN2):c.1695G>T (p.Met565Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M565I variant (also known as c.1695G>T), located in coding exon 5 of the AXIN2 gene, results from a G to T substitution at nucleotide position 1695. The methionine at codon 565 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
