Pathogenic for Capillary malformation-arteriovenous malformation syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002890.3(RASA1):c.2926-14C>G, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RASA1 gene (transcript NM_002890.3) at 14 bases into the intron immediately before coding-DNA position 2926, where C is replaced by G. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has been observed in individual(s) with clinical features of capillary malformation-arteriovenous malformation (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 23 of the RASA1 gene. It does not directly change the encoded amino acid sequence of the RASA1 protein.
