Uncertain significance for Neuromuscular disease caused by qualitative or quantitative defects of dysferlin — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001130987.2(DYSF):c.5831G>A (p.Arg1944Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with glutamine at codon 1905 of the DYSF protein (p.Arg1905Gln). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and glutamine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with DYSF-related conditions. ClinVar contains an entry for this variant (Variation ID: 285259). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
