Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001374385.1(ATP8B1):c.918_919insTTTTCTGTCCTTTGTCCATCAAAACCAAAAGAAGAGTAAT (p.His307delinsPheSerValLeuCysProSerLysProLysGluGluTer), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.His307Phefs*13) in the ATP8B1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ATP8B1 are known to be pathogenic (PMID: 15239083, 22525741). This variant has not been reported in the literature in individuals affected with ATP8B1-related conditions.
