Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001378609.3(OTOGL):c.3363del (p.Cys1122fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the OTOGL gene (transcript NM_001378609.3) at coding-DNA position 3363, deleting one base; at the protein level this means shifts the reading frame starting at cysteine residue 1122, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with OTOGL-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Cys1113Valfs*29) in the OTOGL gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in OTOGL are known to be pathogenic (PMID: 23122586).
