Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5740C>G (p.Leu1914Val), citing Ambry Variant Classification Scheme 2023: The p.L1914V variant (also known as c.5740C>G), located in coding exon 42 of the POLE gene, results from a C to G substitution at nucleotide position 5740. The leucine at codon 1914 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
