This variant lies in the STXBP2 gene (transcript NM_006949.4) at coding-DNA position 1364 through coding-DNA position 1377, deleting 14 bases; at the protein level this means shifts the reading frame starting at glycine residue 455, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the STXBP2 protein in which other variant(s) (p.Gly541Ser) have been determined to be pathogenic (PMID: 20558610, 20798128, 20823128, 24194549). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This sequence change creates a premature translational stop signal (p.Gly455Alafs*120) in the STXBP2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 139 amino acid(s) of the STXBP2 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with STXBP2-related conditions.
