Pathogenic for Myoclonic dystonia 11 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003919.3(SGCE):c.235G>T (p.Glu79Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SGCE gene (transcript NM_003919.3) at coding-DNA position 235, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 79 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has not been reported in the literature in individuals affected with SGCE-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu79*) in the SGCE gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SGCE are known to be pathogenic (PMID: 12821748, 15389977, 17853490, 24297365).
