Uncertain significance — the classification assigned by GeneDx to NM_213599.3(ANO5):c.845A>G (p.Tyr282Cys), citing GeneDx Variant Classification (06012015). This variant lies in the ANO5 gene (transcript NM_213599.3) at coding-DNA position 845, where A is replaced by G; at the protein level this means replaces tyrosine at residue 282 with cysteine — a missense variant. Submitter rationale: The Y282C variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The Y282C variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). This variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is not conserved, and in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
