Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015466.4(PTPN23):c.1906C>T (p.Gln636Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gln636*) in the PTPN23 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PTPN23 are known to be pathogenic (PMID: 29090338, 29899372). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PTPN23-related conditions.
