Pathogenic for Primary ciliary dyskinesia 19 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012472.6(DNAAF11):c.845dup (p.Lys283fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DNAAF11 gene (transcript NM_012472.6) at coding-DNA position 845, duplicating one base; at the protein level this means shifts the reading frame starting at lysine residue 283, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with LRRC6-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Lys283Glufs*12) in the LRRC6 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in LRRC6 are known to be pathogenic (PMID: 23122589).
