Likely pathogenic for Bardet-Biedl syndrome 10 — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_024685.4(BBS10):c.197+1G>T, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BBS10 gene (transcript NM_024685.4) at the canonical splice donor site of the intron immediately after coding-DNA position 197, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: The variant, BBS10 c.197+1G>T is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Five predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 241132 control chromosomes (gnomAD) and has been reported in the literature in an individual affected with Bardet-Biedl Syndrome (Deveault_2011). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as likely pathogenic.
