Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_212482.4(FN1):c.643T>C (p.Cys215Arg), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FN1 gene (transcript NM_212482.4) at coding-DNA position 643, where T is replaced by C; at the protein level this means replaces cysteine at residue 215 with arginine — a missense variant. Submitter rationale: This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 215 of the FN1 protein (p.Cys215Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of spondylometaphyseal dysplasia-corner fracture type (Invitae). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on FN1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
