Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001017420.3(ESCO2):c.1225C>T (p.Gln409Ter), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with ESCO2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln409*) in the ESCO2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ESCO2 are known to be pathogenic (PMID: 15821733, 16380922). For these reasons, this variant has been classified as Pathogenic.
