This variant lies in the AXIN2 gene (transcript NM_004655.4) at coding-DNA position 1800, deleting one base; at the protein level this means shifts the reading frame starting at glycine residue 601, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant has not been reported in the literature in individuals affected with AXIN2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gly601Alafs*88) in the AXIN2 gene. Loss-of-function variants in AXIN2 are known to be pathogenic (PMID: 21416598, 15042511).
