This variant lies in the PALB2 gene (transcript NM_024675.4) at 4 bases upstream of the translation start (5' untranslated region) through coding-DNA position 15, deleting this region; at the protein level this means shifts the reading frame starting at methionine residue 1, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant disrupts a region of the PALB2 protein in which other variant(s) (p.Leu35Pro) have been determined to be pathogenic (PMID: 28319063, 30337689, 31586400). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
