Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the GNAS protein in which other variant(s) (p.Leu179Arg) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant results in the deletion of exon 7 and part of exon 8 (c.531-105_599del) of the GNAS gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in GNAS are known to be pathogenic (PMID: 11784876, 23281139, 23796510, 25802881). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GNAS-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
