Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007325.5(GRIA3):c.1564dup (p.Ser522fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser522Phefs*33) in the GRIA3 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in GRIA3 cause disease. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of early infantile epileptic encephalopathy (internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 2832255). For these reasons, this variant has been classified as Pathogenic.
