This variant lies in the PIGN gene (transcript NM_176787.5) at coding-DNA position 1533, where T is replaced by A; at the protein level this means converts the codon for tyrosine at residue 511 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with PIGN-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr511*) in the PIGN gene.
