This variant lies in the POLD1 gene (transcript NM_002691.4) at coding-DNA position 1137, where G is replaced by T; at the protein level this means replaces glutamine at residue 379 with histidine — a missense variant. Submitter rationale: Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
