Pathogenic for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001277115.2(DNAH11):c.9337-12_9398dup, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DNAH11 gene (transcript NM_001277115.2) at 12 bases into the intron immediately before coding-DNA position 9337 through coding-DNA position 9398, duplicating this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with DNAH11-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ala3134Phefs*8) in the DNAH11 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DNAH11 are known to be pathogenic (PMID: 18022865, 20513915, 22184204).
