Pathogenic for Dystonia 12 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152296.5(ATP1A3):c.2167G>C (p.Ala723Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ATP1A3 gene (transcript NM_152296.5) at coding-DNA position 2167, where G is replaced by C; at the protein level this means replaces alanine at residue 723 with proline — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 723 of the ATP1A3 protein (p.Ala723Pro). For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ATP1A3 protein function. This missense change has been observed in individual(s) with clinical features of developmental and epileptic encephalopathy (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency).
