This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 9667, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 3223 with glutamine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces glutamic acid with glutamine at codon 3250 of the PLEC protein (p.Glu3250Gln). The glutamic acid residue is moderately conserved and there is a small physicochemical difference between glutamic acid and glutamine.
