Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004465.2(FGF10):c.527T>C (p.Met176Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 176 of the FGF10 protein (p.Met176Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with lacrimo-auriculo-dento-digital syndrome (Invitae). It has also been observed to segregate with disease in related individuals. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
