Pathogenic for Hereditary insensitivity to pain with anhidrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002529.4(NTRK1):c.126_127dup (p.His43fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NTRK1 gene (transcript NM_002529.4) at coding-DNA position 126 through coding-DNA position 127, duplicating 2 bases; at the protein level this means shifts the reading frame starting at histidine residue 43, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with NTRK1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.His43Profs*27) in the NTRK1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NTRK1 are known to be pathogenic (PMID: 10982191).
