Pathogenic for Congenital muscular dystrophy due to integrin alpha-7 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002206.3(ITGA7):c.2276del (p.Gln759fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ITGA7 gene (transcript NM_002206.3) at coding-DNA position 2276, deleting one base; at the protein level this means shifts the reading frame starting at glutamine residue 759, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with ITGA7-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln759Argfs*22) in the ITGA7 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ITGA7 are known to be pathogenic (PMID: 9590299).
