NM_000535.7(PMS2):c.1990C>G (p.Leu664Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L664V variant (also known as c.1990C>G), located in coding exon 11 of the PMS2 gene, results from a C to G substitution at nucleotide position 1990. The leucine at codon 664 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
