This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 3142, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1048 with lysine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 1075 of the PLEC protein (p.Glu1075Lys). This variant is present in population databases (rs199523622, gnomAD 0.06%). This variant has not been reported in the literature in individuals affected with PLEC-related conditions.
