Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000781.3(CYP11A1):c.809G>A (p.Trp270Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Trp270*) in the CYP11A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CYP11A1 are known to be pathogenic (PMID: 15507506, 22435390, 27855232, 229968487). This variant has not been reported in the literature in individuals affected with CYP11A1-related conditions. For these reasons, this variant has been classified as Pathogenic.
