Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.8255G>A (p.Arg2752Gln), citing Ambry Variant Classification Scheme 2023: The p.R2752Q variant (also known as c.8255G>A), located in coding exon 24 of the DSP gene, results from a G to A substitution at nucleotide position 8255. The arginine at codon 2752 is replaced by glutamine, an amino acid with highly similar properties. This variant was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
