Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001369268.1(ACAN):c.760G>T (p.Glu254Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu254*) in the ACAN gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ACAN are known to be pathogenic (PMID: 16080123, 24762113). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ACAN-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
