Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000441.2(SLC26A4):c.1545T>A (p.Phe515Leu), citing Invitae Variant Classification Sherloc (09022015): An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. For these reasons, this variant has been classified as Pathogenic. This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 515 of the SLC26A4 protein (p.Phe515Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with non-syndromic recessive deafness (PMID: 34943614). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals.
