Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015629.4(PRPF31):c.3G>C (p.Met1Ile), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Disruption of the initiator codon has been observed in individuals with retinitis pigmentosa (PMID: 28041643; Invitae). This variant is not present in population databases (gnomAD no frequency). This sequence change affects the initiator methionine of the PRPF31 mRNA. The next in-frame methionine is located at codon 59.
