Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004562.3(PRKN):c.844C>T (p.Gln282Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PRKN-related conditions. For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gln282*) in the PRKN gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PRKN are known to be pathogenic (PMID: 10072423, 20301651, 22956510).
