Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000342.4(SLC4A1):c.2659_2662dup (p.Ala888fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SLC4A1 gene (transcript NM_000342.4) at coding-DNA position 2659 through coding-DNA position 2662, duplicating 4 bases; at the protein level this means shifts the reading frame starting at alanine residue 888, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Ala888Glyfs*3) in the SLC4A1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 24 amino acid(s) of the SLC4A1 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autosomal dominant renal tubular acidosis (Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic.
