Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_013432.5(TONSL):c.1A>C (p.Met1Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change affects the initiator methionine of the TONSL mRNA. The next in-frame methionine is located at codon 80. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TONSL-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant disrupts a region of the TONSL protein in which other variant(s) (p.Arg42His) have been observed in individuals with TONSL-related conditions (PMID: 30773278). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
