Pathogenic for Developmental and epileptic encephalopathy, 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001184880.2(PCDH19):c.2266_2273del (p.Lys756fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PCDH19 gene (transcript NM_001184880.2) at coding-DNA position 2266 through coding-DNA position 2273, deleting 8 bases; at the protein level this means shifts the reading frame starting at lysine residue 756, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has not been reported in the literature in individuals affected with PCDH19-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Lys756Profs*8) in the PCDH19 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PCDH19 are known to be pathogenic (PMID: 21053371).
