Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000548.5(TSC2):c.2294C>T (p.Ala765Val), citing Sema4 Curation Guidelines. This variant lies in the TSC2 gene (transcript NM_000548.5) at coding-DNA position 2294, where C is replaced by T; at the protein level this means replaces alanine at residue 765 with valine — a missense variant. Submitter rationale: The TSC2 c.2294C>T (p.A765V) variant has not been reported in the literature to our knowledge. This variant was observed in 2/15434 chromosomes in the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID: 281384). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
