Pathogenic for Nephronophthisis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001128178.3(NPHP1):c.1479dup (p.Leu494fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with NPHP1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Leu550Thrfs*55) in the NPHP1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NPHP1 are known to be pathogenic (PMID: 23559409).
