Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251480 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, c.558C>A has not been observed in individual(s) affected with Galactosemia. A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.556C>T, p.His186Tyr), supporting the critical relevance of codon 186 to GALT protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
