Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001122659.3(EDNRB):c.449A>C (p.His150Pro), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change replaces histidine, which is basic and polar, with proline, which is neutral and non-polar, at codon 150 of the EDNRB protein (p.His150Pro). This missense change has been observed in individual(s) with autosomal dominant Waardenburg syndrome (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt EDNRB protein function. For these reasons, this variant has been classified as Pathogenic.
