NM_002691.4(POLD1):c.2366C>T (p.Pro789Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P789L variant (also known as c.2366C>T), located in coding exon 18 of the POLD1 gene, results from a C to T substitution at nucleotide position 2366. The proline at codon 789 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
