Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006445.4(PRPF8):c.6172_6283del112 (p.Asn2061fs), citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of part of exons 38-39 (c.6174_6285del) of the PRPF8 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in PRPF8 are known to be pathogenic (PMID: 27208204, 31054281, 33946315). This variant is not present in population databases (gnomAD no frequency). A similar copy number variant has been observed in individual(s) with clinical features of retinitis pigmentosa (Invitae). For these reasons, this variant has been classified as Pathogenic.
