Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4753A>G (p.Lys1585Glu), citing Ambry Variant Classification Scheme 2023: The p.K1585E variant (also known as c.4753A>G), located in coding exon 36 of the TSC2 gene, results from an A to G substitution at nucleotide position 4753. The lysine at codon 1585 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
