The c.1378_1381delCTTG variant causes a frameshift starting with codon Leucine 460, changes this amino acid to an Isoleucine residue, and creates a premature Stop codon at position 131 of the new reading frame, denoted p.Leu460IlefsX131. This variant is predicted to cause loss of normal protein function through protein truncation with the last 304 amino acid resides replaced by 130 incorrect amino acid residues. The c.1378_1381delCTTG variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.1378_1381delCTTG as a pathogenic variant.
