It does not directly change the encoded amino acid sequence of the SCN1A protein. However, this sequence change falls within a region encompassing a cryptic exon in the SCN1A gene, in which variants have been shown to alter splicing (PMID: 30526861, 34107977). This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with clinical features of SCN1A-related conditions (Invitae). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing.
