Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_015450.3(POT1):c.1250C>T (p.Ser417Leu), citing Ambry Variant Classification Scheme 2023: The p.S417L variant (also known as c.1250C>T), located in coding exon 10 of the POT1 gene, results from a C to T substitution at nucleotide position 1250. The serine at codon 417 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
