Pathogenic for Metachromatic leukodystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000487.6(ARSA):c.854+1G>T, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 4 of the ARSA gene. RNA analysis indicates that this variant induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). Disruption of this splice site has been observed in individual(s) with metachromatic leukodystrophy (PMID: 7833949). ClinVar contains an entry for this variant (Variation ID: 280764). Studies have shown that disruption of this splice site is associated with exon skipping, which introduces a premature termination codon (PMID: 7833949). The resulting mRNA is expected to undergo nonsense-mediated decay. For these reasons, this variant has been classified as Pathogenic.
