Pathogenic — the classification assigned by GeneDx to NM_012330.4(KAT6B):c.5101C>T (p.Gln1701Ter), citing GeneDx Variant Classification (06012015). This variant lies in the KAT6B gene (transcript NM_012330.4) at coding-DNA position 5101, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 1701 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q1701X pathogenic variant in the KAT6B gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function through protein truncation. The Q1701X variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret Q1701X as a pathogenic variant.
